Target intelligence / Profile preview
Nail keratin is a structural protein forming the main component of the nail plate, providing rigidity and mechanical protection. It is highly cross-linked by disulfide bonds, making it insoluble and resistant to proteolytic enzymes, which contributes to the formidable barrier property of the nail. Fungal cell structures, particularly the cell wall, are composed of chitin, β-glucans, mannans, and other glycoproteins that provide both structural integrity and dynamic adaptability, and are essential for fungal growth, morphogenesis, and pathogenicity. In onychomycosis, dermatophyte fungi (such as Trichophyton species) invade the keratinized nail plate; their ability to digest keratin and remodel their cell walls plays a central role in infection and virulence. Treatments target fungal cell wall biosynthesis and attempt to overcome the nail's barrier properties to deliver drugs effectively. However, this functional designation ("Nail Keratin and Fungal Cell Structures") is not a single molecular target but rather describes relevant host and pathogen biology in fungal nail disease. Critical note: This entry does not refer to a conventional drug target in the sense of molecular pharmacology but rather an anatomical and pathogen structure relevant to disease mechanism and therapy resistance.
Antifungals: Inhibit fungal cell wall synthesis, disrupt fungal membrane synthesis, or increase nail plate permeability to enable drug access
