Keratin 74 is a type II intermediate filament protein expressed exclusively in the differentiating (keratinizing) epithelial cells of the Huxley layer of the inner root sheath of hair follicles. As a keratin, its primary role is to provide structural stability to epithelial cells, particularly in the context of hair formation. Mutations in the KRT74 gene are associated with inherited hair disorders like monilethrix and hypotrichosis, which result in fragile or sparse hair. Keratin 74 is not considered a receptor, enzyme, or major therapeutic target, but its function is crucial for normal hair follicle differentiation and integrity. There is no indication of druggability, established pharmacological targeting, or major safety concerns related to KRT74 in clinical medicine.
