Overview

Keratin-associated protein 1-1 (KRTAP1-1) is a member of the high-sulfur keratin-associated protein (KAP) family, which forms the matrix enveloping hair keratin intermediate filaments and is critical for the structure and properties of hair fibers. KRTAP1-1 is embedded in the hair cortex where it participates in the assembly and stabilization of keratin bundles, primarily through extensive disulfide bond cross-linking due to abundant cysteine residues. This results in a rigid and resistant hair shaft. KAP family proteins have unique family-specific amino and carboxyl terminal regions and KRTAP1-1 is localized on human chromosome 17q21.2. KRTAP1-1 does not appear to be a conventional therapeutic target (e.g., receptor, enzyme, transporter) but rather a structural protein important in hair biology. However, genetic variation in this gene has been associated with response to antidepressant therapy, suggesting a potential role as a biomarker in pharmacogenomic research.
