Overview

Keratin-associated protein 1-3 (KRTAP1-3) is a member of the keratin-associated protein (KAP) family, specifically classified among the high-sulfur KAPs[1][6]. These proteins contribute to the structural matrix of keratin intermediate filaments in hair fibers, interacting closely with hair keratins via extensive disulfide cross-linking, predominantly through cysteine residues. This association imparts rigidity and resilience to the hair shaft by forming a robust interfilamentous matrix[1][9]. KRTAP1-3 is located in a cluster of KAP genes at chromosome 17q12-q21[1][7], and its primary role is structural, being essential for proper hair fiber formation and properties. Although variants are associated with hair structure, KRTAP1-3 is not a receptor, enzyme, transporter, or other typical drug target, and there are no known drugs or therapeutic safety concerns related to this protein[1][2][7][9]. Choroid cancer is referenced as a disease association, but precise pathogenetic roles are unclear[1].
