Keratin-associated protein 12-1 (KRTAP12-1) is a structural protein belonging to the keratin-associated protein (KAP) family, specifically the high-sulfur group, based on its amino acid composition[3][6]. KRTAP12-1 is predominantly expressed in the hair cortex, where it acts as an interfilamentous matrix protein embedding hair keratin intermediate filaments. This is achieved through extensive disulfide bond cross-linking via the abundant cysteine residues, which is crucial for the mechanical strength, rigidity, and resistance of the hair shaft[3][6]. KRTAP12-1 and related KAPs are classified mainly by their high cysteine (sulfur) content and play an essential role in forming the structural matrix that supports keratin filaments in hair fibers[6]. There is no evidence that KRTAP12-1 is directly involved in signaling, metabolic, or receptor-mediated pathways, nor is it currently considered a therapeutic target; it is primarily of interest in the context of hair structure, development, and potentially genetic variation related to hair traits[3][6][2].
