Keratin-associated protein 12-4 (KRTAP12-4) is a structural protein encoded by the KRTAP12-4 gene, located on human chromosome 21 (21q22.1), and is a member of the high sulfur keratin-associated protein family[4][2]. KRTAP12-4 is expressed in the hair cortex, where it binds with keratin intermediate filaments to form a rigid and resilient matrix that gives hair shaft its strength and stability through extensive disulfide bond cross-linking of cysteine residues[4][2]. This protein, together with other high sulfur KRTAPs, plays a critical role in determining the physical properties of hair fibers such as strength, rigidity, and flexibility[2]. While no direct pathophysiological role in disease has been established, variation within the KRTAP family can contribute to differences in hair properties among mammals[2][4][6]. There are no reports of known drugs interacting with, or acting through, KRTAP12-4, and it is not being investigated as a therapeutic target or biomarker. Its major biological importance is limited to the process of keratinization and hair formation[4][2][6].
