Overview

Keratin-associated protein 2-2 (KRTAP2-2) is a structural protein predominantly expressed in the hair shaft cortex, belonging to a multigene family of keratin-associated proteins (KAPs) that are major components of the hair and wool fiber matrix[2][4][5]. KRTAP2-2 cross-links with hair keratin intermediate filaments via disulfide bonds, contributing to the strength and rigidity of hair fibers[2][4]. KRTAP2-2 is classified as a high-sulfur or high glycine-tyrosine KAP, characterized by high levels of cysteine, glycine, and tyrosine residues that facilitate cross-linking and affect hair structure and mechanical properties[2][3][4]. It is not considered a conventional therapeutic target such as a receptor, enzyme, or transporter, but genetic variation in KRTAP2-2 may modify hair properties and be relevant to hair biology research and certain hair pathologies[4][5].
