Keratin-associated protein 20-3 (KRTAP20-3) is a member of the keratin-associated protein (KAP) family of structural proteins important in hair and wool fiber structure. It is part of the KAP family, which are small proteins (typically 10–30 kDa) that play a critical role in the structure and properties of wool and hair fibers in mammals. KAPs form the protein matrix that embeds keratin intermediate filaments (KIFs), the major structural elements of wool and hair. They are characterized by being rich in cysteine, glycine, or tyrosine; high glycine and tyrosine KAPs (HGT-KAPs) like KRTAP20-3 can mediate cross-linking between fibers and contribute to fiber strength and pliability through both sulfur- and aromatic ring-mediated interactions. Genetically, KRTAP proteins are encoded by single-exon genes. In sheep and other livestock, KRTAP genes, including those in the KRTAP20 family, affect important wool traits. For example, variants in closely related KRTAP20-1 show associations with mean fiber diameter and fleece yield. KRTAP20-3 itself is primarily studied for its impact on fiber and fleece traits in animals, not as a therapeutic or diagnostic target in human medicine. There are no known drugs, established biomarkers, or disease indications associated with KRTAP20-3 in humans or animals. In summary, KRTAP20-3 and its family members are important for the structural biology of mammalian hair and wool fiber but have no established role as classical therapeutic targets, disease biomarkers, or drug-interacting proteins.
