Overview

Keratin-associated protein 4-6 is a member of the ultrahigh sulfur subfamily of the keratin-associated protein (KAP) family, which plays an essential role in the structural formation and mechanical stabilization of hair fibers. In the hair cortex, keratin intermediate filaments are embedded in a matrix composed of KAPs, including KRTAP4-6, which interact with keratins through extensive disulfide bonds facilitated by high cysteine content. These interactions are critical for creating strong, rigid, and resilient hair shafts. KRTAP4-6 is thus involved in the formation and maintenance of hair structure, but is not considered a classical therapeutic target such as a receptor, enzyme, transporter, or transcription factor. The gene is localized to chromosome 17q21 in humans and is one of many clustered KAP family members responsible for hair morphology variation across mammals[2][3][4][5]. There is no evidence of drug interactions, biomarker roles, or notable safety concerns related to KRTAP4-6.
