Overview

Keratin-associated protein 5-1 (KRTAP5-1) is a member of the keratin-associated protein (KAP) family, which are essential structural proteins in hair and nails[1][3][4]. KRTAP5-1 is present in the hair cortex, where it contributes to the formation of a rigid and resistant hair shaft by cross-linking extensively with hair keratin intermediate filaments via disulfide bonds, thanks to its abundant cysteine residues[1][3]. These matrix proteins are particularly high in sulfur, a property that allows the formation of stable, resilient structures. KRTAP5-1 is not regarded as a classical drug target (e.g., enzyme, receptor, transporter) and there is no current evidence for its direct involvement in disease or for use as a biomarker or safety concern. However, the related protein KRTAP5-5 has been implicated in cancer cell cytoskeletal dynamics, suggesting that some keratin-associated proteins may play more active cellular roles[2].
