Overview

Keratin-associated protein 5-8 is a member of the ultra-high sulfur keratin-associated proteins found primarily in the hair cortex and skin epidermis. These proteins form an interfilamentous matrix around hair keratin intermediate filaments, contributing to the mechanical resilience and rigidity of hair shafts through extensive disulfide bond cross-linking. They function as structural proteins that help determine hair fiber properties and integrity, but do not have enzymatic or signaling roles. KRTAP5-8 is predominantly expressed in hair follicles and is predicted to be localized in the cytosol as part of the structural matrix. While related members in the KRTAP family can influence cellular processes through their structural roles, KRTAP5-8 is not currently considered a therapeutic target, nor are there any drugs, clinical biomarkers, or safety issues linked to modulation of its function. Its relevance is limited to supporting hair and epidermal structure, and its deficiency or mutation could hypothetically contribute to hair fiber anomalies, but no direct disease association is documented in available databases and literature.
