Overview

Keratin-associated protein 6-2 (KRTAP6-2) is a structural protein primarily expressed in hair and other keratinized tissues. It is found in the matrix surrounding keratin intermediate filaments in the hair cortex, enabling the formation of a rigid and resistant hair shaft through extensive disulfide bond cross-linking with hair keratins. KRTAP6-2 is part of a broadly conserved mammalian gene family, classified among the high-sulfur or high-glycine-tyrosine keratin-associated proteins involved in keratinization. While variation in KRTAP genes may impact hair properties and has been linked in some studies to traits such as hair structure, there is no evidence that KRTAP6-2 serves as a therapeutic target, and it is not known to interact with any drugs or be a recognized biomarker. KRTAP6-2 is located on human chromosome 21 and is predicted to localize in the cytosol. The KRTAP family plays a significant role in the mechanical strength and structure of hair through its cross-linking capability with keratin fibers. Variations in KRTAP genes, particularly in sheep and to a lesser extent in humans, can result in differences in hair and wool properties, but clinical disease associations are rare and likely indirect. KRTAP6-2 is not a typical drug target (such as a receptor, enzyme, transporter, etc.) and is not currently targeted in therapeutic interventions.
