Keratinized stratified squamous epithelium is a type of tissue found in the epidermis and certain oral regions, composed of layers of squamous cells (keratinocytes) whose outermost layer undergoes keratinization (cornification). This results in a robust, waterproof barrier that protects against mechanical abrasion, desiccation, and pathogens. Keratinized tissue provides regional resistance to physical stress and is distinguished by a multi-layered cellular architecture with increased keratin protein content in the outer layers. The process of keratinization involves terminal differentiation, loss of cellular organelles, and formation of a rigid, protein-rich cell envelope. Disease associations center on keratin gene mutations or failures in proper keratinization, affecting skin and mucosal integrity.
