Keratinized tissue is a specialized form of stratified squamous epithelium characterized by the presence of a cornified layer (stratum corneum) composed of dead cells filled with keratin filaments. This tissue serves as the body's primary physical barrier, protecting against mechanical trauma, chemical insults, microbial invasion, and water loss (StatPearls, 2023). It is predominantly found in the epidermis of the skin but also occurs in specialized areas like the gingiva and hard palate. While keratinized tissue is a histological classification rather than a specific molecular target, it is the physiological site for numerous dermatological interventions. Drugs such as keratolytics (e.g., salicylic acid) work by breaking down the intercellular matrix in this tissue to promote shedding, while retinoids influence the underlying cellular differentiation processes that produce the keratinized layer (NIH, 2022). Disorders of keratinization, such as psoriasis and ichthyosis, involve the abnormal accumulation or formation of this tissue, making its regulation a key focus in clinical dermatology. Therapeutic strategies often aim to restore the balance of keratinization to maintain skin integrity and function (PubMed, 2021).
