Keratinocyte differentiation-associated protein (KRTDAP) is a small, secreted regulatory protein expressed predominantly by differentiating keratinocytes in the granular layer of stratified epithelia, especially the skin[1][2][3][6][10]. It appears to act as a soluble regulator of keratinocyte differentiation, cornification (the final stage of skin cell differentiation), and/or desquamation, and has roles in embryonic skin morphogenesis[1][2][3][6]. The gene is tightly regulated during epidermal development, and altered KRTDAP expression is observed in several epithelial pathologies, including squamous cell carcinoma, head and neck squamous cell carcinoma, mesothelioma, bladder cancer, inflammatory bowel disease (type 13), and white sponge nevus[1][2][4]. Downregulation is particularly associated with more aggressive head and neck squamous cell carcinoma and its expression is explored as a diagnostic and prognostic biomarker in these settings[2][4]. There are no approved drugs known to directly interact with KRTDAP, nor evidence that it serves as a conventional therapeutic target (e.g., enzyme, receptor, transporter).
