Overview

Keratinocyte differentiation factor 1 (KDF1) is a cytoplasmic protein encoded by the KDF1 gene, primarily involved in the regulation of epidermal development, specifically promoting cellular differentiation and inhibiting excessive proliferation in keratinocytes. KDF1 participates in critical biological processes such as skin and epidermal formation, with loss-of-function variants causing ectodermal dysplasia syndromes in humans and mice. Overexpression of KDF1 has been linked to the progression, proliferation, and metastasis of certain cancers (notably epithelial ovarian and cervical cancer), where it contributes to cancer cell migration and invasion through pathways like Wnt/β-catenin and PI3K/AKT, and may serve as a prognostic biomarker or therapeutic target in these settings. KDF1 also physically interacts with proteins such as E-cadherin and IκB kinase α, influencing cell-cell adhesion and signaling cascades relevant to development and cancer biology. No specific drugs directly targeting KDF1 are currently reported, nor are there established clinical safety concerns, but perturbation of KDF1 function may affect epidermal integrity given its developmental role.
