Overview

Keratinocyte proliferation and differentiation refer to the tightly regulated processes by which epidermal keratinocytes divide and then mature as they migrate outward from the basal layer to form the protective stratified layers of the skin. In the basal layer, keratinocytes undergo periodic cell division (proliferation), then arrest growth, lose proliferative capacity, and express a specialized terminally differentiated phenotype (differentiation)[2][3]. These processes are crucial for epidermal homeostasis, wound healing, and defense against environmental insults like ultraviolet light. Dysregulation can result in hyperproliferative disorders such as psoriasis, or contribute to skin carcinogenesis. Agents that modulate these processes are under investigation for chemoprevention and the treatment of inflammatory skin diseases[1][3]. Numerous molecular pathways—including cytokines, cell cycle regulators, and differentiation markers—govern these events, with many potential drug targets embedded within these pathways, but "keratinocyte proliferation and differentiation" itself is not a standalone molecular target[1][2][3][5][6][7].
