Overview

KH domain-containing, RNA-binding, signal transduction-associated protein 3 (KHDRBS3) is an RNA-binding protein that regulates alternative splicing events and influences mRNA splice site selection and exon inclusion[1][2][3]. It recognizes and binds specific RNA motifs, modulating the expression and splice variants of key genes involved in processes such as cell proliferation, neuronal synaptic function, spermatogenesis, and sarcomere organization in cardiac muscle[1][2][3]. Overexpression or altered regulation of KHDRBS3 has been implicated in several cancers (including medulloblastoma, breast, gastric, and prostate cancer) where it fosters cancer cell stemness and survival, most notably by modulating CD44 splicing[2][3]. KHDRBS3 also interacts with other splicing factors (e.g., Sam68) and can be regulated by cellular kinases such as PTK6[1]. In the heart, increased KHDRBS3 expression is observed in failing myocardium, where it binds transcripts relevant to sarcomere integrity[3]. No drugs are reported to directly target KHDRBS3, but its role in alternative splicing and tumor progression makes it a candidate for therapeutic exploration[2][3].
