KICSTOR subunit 2 (KICS2), encoded by the C12orf66 gene, is a key component of the KICSTOR complex, which comprises four proteins. This complex localizes to the lysosomal membrane and recruits GATOR1, a negative regulator, to suppress mTORC1 signaling under conditions of amino acid or glucose deprivation. Proper mTORC1 inhibition is necessary for controlled cell growth and proliferation. Loss of KICS2 disrupts nutrient sensing and can lead to constitutive mTORC1 activation. KICS2 is widely expressed, with higher levels in endocrine, lymphoid tissues, and early developmental stages, and altered expression in various cancers and neurodevelopmental disorders. KICS2 is not currently a direct drug target, but its pathway (mTOR signaling) is pharmacologically targeted in several disease contexts[1][2][6].
