Overview

Killer cell immunoglobulin-like receptor 3DL3 (KIR3DL3) is an inhibitory receptor primarily expressed on subsets of T cells, especially γδ T cells and CD8+ T cells, and terminally differentiated effector memory NK cells. It interacts with HHLA2, a B7 family member, mediating co-inhibition of immune responses. KIR3DL3 acts as an immune checkpoint, and blockade of the KIR3DL3-HHLA2 interaction can enhance anti-tumor immunity. The gene is highly polymorphic, affecting expression levels, making it relevant to personalized medicine approaches.
