KIR2DL3 is an inhibitory receptor expressed on NK cells and some T cells. It binds to specific HLA-C alleles (Cw1, Cw3, Cw7) and inhibits NK cell activation via ITIM signaling, playing a crucial role in regulating immune responses and preventing autoimmunity. Polymorphisms in KIR2DL3 and its ligands are associated with susceptibility to various diseases, including viral infections, cancer, and autoimmune conditions. Therapeutic antibodies targeting KIRs, such as Lirilumab, are under investigation for cancer treatment.
Upon ligand binding, ITIM motifs are phosphorylated, recruiting phosphatases (SHP‐1/SHP‐2) that dephosphorylate key signaling proteins, inhibiting cellular activation/cytotoxicity against target cells expressing compatible MHC class I molecules.
