Overview

Killer cell lectin-like receptor C1 (NKG2A) is an inhibitory receptor expressed on natural killer (NK) cells and some cytotoxic T lymphocytes. It forms a heterodimer with CD94 and recognizes HLA-E presenting peptides derived from the leader sequences of MHC class I molecules. This interaction delivers inhibitory signals that suppress NK cell activation against normal "self" targets. NKG2A is a therapeutic target in cancer immunotherapy, with antibodies like monalizumab being developed to block its inhibitory function and enhance NK cell-mediated killing of tumor cells.
