NKG2A is an inhibitory immune checkpoint receptor expressed on NK cells and CD8+ T cells. It forms a heterodimer with CD94 and binds to HLA-E, a non-classical MHC class I molecule. This interaction delivers inhibitory signals, preventing the activation of cytotoxic lymphocytes. Blocking this interaction enhances anti-tumor immunity and is a target for cancer immunotherapy.
NKG2A antagonists block the interaction between NKG2A and HLA-E, thereby restoring the cytotoxic activity of NK and CD8+ T cells.
