Overview

Killer cell lectin-like receptor subfamily G member 1 (KLRG1) is a lymphocyte co-inhibitory receptor, primarily expressed on late-differentiated effector and effector memory CD8+ T cells and natural killer (NK) cells. It acts as an inhibitory receptor that regulates the cytolytic activity of NK cells and modulates both innate and adaptive immune responses by inhibiting proliferation of activated T lymphocytes and reducing NK cell effector function. KLRG1 ligands include E-cadherin and N-cadherin. It is considered a marker of cellular senescence or terminal differentiation in T cells.

Ligand engagement leads to ITIM phosphorylation and recruitment of phosphatases (SHP-2, SHIP-1) to mediate inhibitory signaling.
