Overview

Killer immunoglobulin-like receptor 2DL1 (KIR2DL1) is a type I transmembrane glycoprotein expressed on natural killer (NK) cells and some T cell subsets. It regulates NK cell activity by interacting with MHC-I molecules, specifically HLA-C alleles bearing the C2 epitope. Upon ligand binding, KIR2DL1 transduces an inhibitory signal that suppresses NK cell cytotoxicity, preventing lysis of healthy self-cells. Variations in the *KIR* gene cluster are associated with susceptibility or resistance to infections, autoimmune diseases, cancer progression, and outcomes in transplantation settings.
