Overview

KIRREL1 (Kin of IRRE-like protein 1), commonly called NEPH1, is a transmembrane cell adhesion molecule belonging to the immunoglobulin superfamily. It is primarily expressed in kidney podocytes, maintaining the structure of the glomerular filtration barrier and podocyte foot processes via specialized cell-cell junctions called the slit diaphragm. KIRREL1 interacts with cytosolic proteins such as podocin and nephrin and is essential for selective filtration of blood. It also modulates the Hippo signaling pathway by recruiting SAV1 to sites of cell-cell contact and acting as a feedback regulator to inhibit YAP/TAZ-mediated proliferation. Dysregulation or mutation of KIRREL1 is associated with nephrotic syndromes and may play a role in cancer by impacting Hippo signaling[1][2][3][6].
