Overview

"Kinase pathways" refers to the intricate signaling networks orchestrated by kinases, a vast family of enzymes responsible for the transfer of phosphate groups (phosphorylation) to target proteins. This post-translational modification activates or deactivates proteins, enabling rapid and reversible regulation of virtually all cellular processes, including cell proliferation, survival, differentiation, metabolism, and immune responses. Dysregulation of these pathways is a hallmark of many diseases, particularly cancer, and constitutes a major therapeutic strategy via kinase inhibitors. However, this is not a singular molecular target but rather a collective label for any signaling cascade propagated primarily by kinases.

Mechanism of action

The dominant mechanism is inhibition of kinase activity, typically either ATP-competitive inhibition (blocking the kinase catalytic site) or allosteric inhibition, with some monoclonal antibodies blocking extracellular portions of receptor kinases. Multi-kinase or pathway targeting—blocking several signaling nodes simultaneously—is employed to overcome redundancy and resistance.
