Overview

Kinesin family member 19A (KIF19A) is a specialized motor protein belonging to the kinesin-8 subfamily, primarily recognized for its essential role in regulating the length of motile cilia. Unlike most kinesins that facilitate intracellular transport or mitosis, KIF19A localizes to the distal tips of cilia, where it utilizes its ATP-dependent microtubule-depolymerizing activity to maintain optimal ciliary length. This function is critical for generating the fluid flow necessary for physiological processes in the brain ventricles, respiratory tract, and fallopian tubes. Genetic deficiency or dysfunction of KIF19A leads to abnormally elongated cilia, which are unable to beat effectively, resulting in clinical phenotypes such as hydrocephalus and female infertility. In the context of oncology, KIF19A has been identified as a potential promoter of cancer cell invasion and metastasis, with its overexpression correlating with poor prognosis in several malignancies. While no drugs specifically targeting KIF19A are currently approved, it is frequently used as a selectivity target in the development of inhibitors for related kinesins like KIF18A, and it remains a subject of interest for therapeutic intervention in metastatic disease and ciliopathies.
