KIF20A is a mitotic kinesin essential for proper cell division. It plays critical roles in microtubule bundle formation, midbody abscission during cytokinesis, and regulation of cytokinesis through recruitment of key proteins like PLK1. Overexpression or dysregulation of KIF20A has been observed in various cancers, making it a promising anti-cancer drug target. The L6 loop within the N-terminal motor domain contains an unusually long insertion compared to other kinesins, modulating both structural conformation and functional properties and offering a potential druggable site.
