Kinesin family member 20B (KIF20B), also known as MPHOSPH1 or M-phase phosphoprotein 1, is a plus-end-directed kinesin-6 family motor protein crucial for cytokinesis, the final step of cell division. It possesses ATPase activity, drives microtubule-dependent movement, and is phosphorylated during the G2/M cell cycle transition, facilitating proper cell division. KIF20B is overexpressed in various cancers and its upregulation is linked to tumorigenesis, advanced tumor stage, metastasis, and poor prognosis. Functional inhibition of KIF20B results in failed cytokinesis, mitotic arrest, and apoptosis, highlighting it as a promising therapeutic target in oncology. High KIF20B expression can also serve as a tumor biomarker. Its complex with partners like PRC1 is implicated in cancer cell survival and proliferation, especially in genitourinary and hepatocellular malignancies. No approved small molecules directly target KIF20B, but experimental gene silencing and combination approaches enhance antitumor efficacy in preclinical models.
Inhibition leads to mitotic arrest, apoptosis, inhibition of cellular proliferation, and increased sensitivity to mitotic inhibitors (e.g., taxol)
