Kinesin family member 6 (KIF6) is a **molecular motor protein** belonging to the kinesin-9 branch of the kinesin superfamily[1][2][3]. It is essential for **intracellular transport** along microtubules and has a highly specific function in the formation and maintenance of **motile cilia**, particularly those found on brain ependymal cells, which regulate **cerebrospinal fluid flow**. Loss-of-function mutations in KIF6 are linked to **neurodevelopmental disorders**, notably **hydrocephalus and intellectual disability** due to disruption of cilia-mediated fluid movement in the brain's ventricular system[1][2]. KIF6 operates as a plus-end-directed, ATP-driven processive motor, distinct from its close relative KIF9, and is autoinhibited by its C-terminal tail. Unlike KIF9, KIF6 plays a direct role in ciliary cargo transport and coordinated ciliary movement, and its deficiency leads to severe hydrocephalus and reduced ependymal ciliation in animal models[2]. While proposed as a cardiovascular risk locus, broader population studies have not confirmed such a role for KIF6 in heart disease[1]. No drugs currently target KIF6, nor is it an established therapeutic target[3].
