Overview

Kinesin light chain 3 (KLC3) is a microtubule-associated motor protein subunit encoded by the KLC3 gene in humans[1][2][5]. As a member of the kinesin light chain family, KLC3 participates in intracellular cargo transport as part of the heterotetrameric kinesin-1 complex, which includes two kinesin heavy chains and two light chains[1][2][3][4][5]. The primary role of kinesin light chains is to bind specific cargoes and regulate the motor activity of the heavy chains[1][2][3][5]. KLC3 is distinguished from other kinesin light chains by its testis-specific expression, particularly in round and elongating spermatids, suggesting a specialized function during spermiogenesis in the development of the sperm tail midpiece and mitochondrial sheath[2][3][5]. While general kinesin proteins are critical for neuronal and vesicular transport, current evidence does not indicate that KLC3 functions as a direct therapeutic target, receptor, enzyme, or transporter for pharmacological intervention[2][3][5]. KLC3 interacts with kinesin heavy chains using conserved heptad repeat (HR) and tetratricopeptide repeat (TPR) domains that facilitate protein-protein interactions and complex assembly[3]. No specific drugs, diagnostic biomarkers, or recognized safety concerns are directly linked to KLC3 according to current data[2][3][5].
