Kinesin-like protein KIF11 (Eg5)

Target
Eg5
Molecular classification
Motor protein, Kinesin superfamily
01

Overview

Kinesin-like protein KIF11 (Eg5) is a motor protein essential for mitosis in human cells. It establishes bipolar spindle formation during cell division by cross-linking anti-parallel microtubules. Inhibition or loss of function leads to monopolar spindles and cell cycle arrest at mitosis, making it an active anticancer drug target. Small-molecule inhibitors like GSK‑1 trap Eg5 on the microtubule lattice, blocking its function.

Mechanism of action

Inhibition of KIF11 traps Eg5 on the microtubule lattice, stabilizing an "ATP-like" conformation without nucleotide bound, blocking its function allosterically, leading to failed bipolar spindle formation and cell cycle arrest.
