Overview

Kirsten rat sarcoma viral oncogene homolog (KRAS) is a gene encoding the K-Ras protein, a small GTPase that plays a central role in cell signaling pathways regulating proliferation, differentiation, and survival. KRAS mutations, particularly at codons 12, 13, and 61, lock KRAS in its active (GTP-bound) state, leading to continuous activation of proliferative and survival signaling pathways independent of external stimuli, driving uncontrolled cell division and tumorigenesis. Specific inhibitors have been developed targeting the KRAS(G12C) mutant.
