Overview

The KRAS G13D mutation is a missense mutation at codon 13 of the KRAS gene, where glycine (G) is replaced by aspartic acid (D). This alteration leads to constitutive activation of the KRAS protein, impacting cell signaling pathways involved in proliferation, differentiation, and survival. Tumors with the G13D mutation may exhibit intermediate sensitivity/resistance profiles when treated with anti-EGFR monoclonal antibodies. Direct therapeutic targeting remains challenging.
