Overview

KRAS (Kirsten rat sarcoma viral oncogene homolog) is a gene encoding the K-Ras protein, a small GTPase involved in cellular signal transduction. It acts as a molecular switch cycling between active (GTP-bound) and inactive (GDP-bound) states, relaying signals from cell surface receptors to intracellular pathways that control proliferation, differentiation, and survival. Mutations in KRAS are common oncogenic drivers, particularly in pancreatic, colorectal, and lung cancers, leading to constitutive activation and uncontrolled cell growth. Therapeutic targeting is challenging due to structural features and resistance mechanisms, but covalent inhibitors and combination therapies are being explored.
