KLHDC7B divergent transcript (KLHDC7B-DT) is a long non-coding RNA (lncRNA) located at chromosome 22q13.33, transcribed divergently from the KLHDC7B locus[4]. It is upregulated in multiple malignancies, including pancreatic ductal adenocarcinoma and various other cancers, and has also been found differentially expressed in psoriasis lesions[6]. KLHDC7B-DT directly binds to the IL-6 promoter, induces chromatin opening at this locus, and promotes IL-6 transcription and secretion, which triggers autocrine and paracrine STAT3 signaling. These changes foster a tumor-promoting microenvironment by enhancing cancer cell proliferation, migration, invasion, and polarization of macrophages to the M2 phenotype. KLHDC7B-DT is not a protein-coding receptor, enzyme, or transporter, and has no known interacting drugs or established therapeutic targeting mechanisms. It is, however, a potential biomarker and modulator of disease biology, especially in the context of cancer progression and immune environment[1][3][6].
