KN motif and ankyrin repeat domain-containing protein 1 (KANK1) is an adapter protein found in the cytoplasm and nucleus, characterized by coiled-coil and ankyrin repeat domains, and plays a central role in organizing and regulating the actin and microtubule cytoskeletons during cell morphogenesis[1][2][4]. KANK1 negatively regulates cell migration by inhibiting RhoA activation and modulating the binding of signaling molecules such as IRSp53 and 14-3-3, crucial for actin polymerization and formation of cell protrusions[1][2]. It also participates in beta-catenin-dependent transcription when shuttled to the nucleus[1][2][4]. KANK1 acts as a candidate tumor suppressor in renal cell carcinoma and mutations are associated with cerebral palsy spastic quadriplegic type 2 and some myeloproliferative disorders[2][5]. It is not considered a classic therapeutic target such as a receptor or enzyme[2][4].
