Overview

KN motif and ankyrin repeat domains 3 (KANK3) is a member of the KANK protein family, characterized by an N-terminal coiled-coil KN motif and C-terminal ankyrin repeat domains. KANK3 is predicted to regulate the polymerization of actin filaments, thus modulating cytoskeletal organization and cell motility; it acts mainly via negative regulation of actin stress fiber formation and inhibition of RhoA activity, similar to other KANK family proteins. KANK3 is expressed in the cytoplasm and cytoskeleton across multiple tissues. Mutations or dysregulation in KANK family genes are linked to diseases such as cancer and neurodevelopmental disorders, but KANK3 is not established as a therapeutic target nor are there drugs that directly act on it. If more detailed mechanistic or disease information becomes available, a reassessment of its therapeutic target status may be warranted.
