Overview

Krüppel-like factor 9 (KLF9) is a zinc finger transcription factor encoded by the KLF9 gene on human chromosome 9q13, featuring three C-terminal C2H2 zinc fingers that bind GC-rich promoter regions to activate or repress target genes depending on context, such as single versus tandem GC boxes. It functions in diverse processes including circadian rhythm regulation in epidermis via cortisol-dependent expression, antiproliferative effects in keratinocytes, and inhibition of neurite outgrowth in hippocampal neurons partly through repression of cAMP signaling pathway genes like protein kinase A. KLF9 promotes differentiation in epidermal and uterine cells while repressing proliferation, and it responds to oxidative stress by sensitizing cells to reactive oxygen species. In disease, KLF9 acts as a tumor suppressor in cancers like hepatocellular carcinoma by maintaining redox homeostasis and inhibiting inflammation, and it influences immune cell infiltration in the tumor microenvironment; its suppression via miRNAs or lncRNAs is common in neoplasia. Additionally, KLF9 contributes to leukocyte development, hematopoiesis, and female reproductive pathologies. No approved drugs directly target KLF9, limiting its current therapeutic exploration.
