KRAS G12V is a specific point mutation in the KRAS gene, where glycine (G) at position 12 is replaced by valine (V). This mutation results in a constitutively active form of the KRAS protein, leading to continuous downstream signaling through pathways such as RAF/MEK/ERK. It is frequently found in NSCLC, colorectal cancer, and pancreatic ductal adenocarcinoma and is associated with poor prognosis and resistance to EGFR-targeted therapies. There are no FDA-approved selective inhibitors specifically targeting the KRAS G12V mutant.
