KRT19 interacting long noncoding RNA in hepatocellular carcinoma (KILH) is a long noncoding RNA predominantly expressed in KRT19-positive hepatocellular carcinoma cells. KILH binds specifically to KRT19 protein, inhibits its phosphorylation at the Ser35 residue, and enhances KRT19’s interaction with β-catenin, facilitating the nuclear translocation and activation of the Notch1 signaling pathway. Functionally, KILH promotes HCC cell proliferation and metastasis and is upregulated in HCC tissues compared to non-tumor liver tissues, making it a strong candidate for therapeutic targeting and prognostic biomarker in liver cancer. Knockdown of KILH suppresses HCC tumor growth and metastasis in both in vitro and in vivo models.
Drugs targeting KILH would theoretically act by inhibiting its expression or disrupting its interaction with KRT19, thereby reducing Notch1 signaling and decreasing cell proliferation/metastasis
