Krueppel-like factor 12 is a zinc finger transcription factor in the Krueppel-like factor family, characterized by three C2H2-type zinc finger motifs at the carboxy-terminal and a variable amino-terminal regulatory domain. It binds GC-rich DNA sequences to regulate gene expression, notably repressing the AP-2 alpha gene by binding to its promoter with the aid of the corepressor CtBP1. KLF12 is involved in cell proliferation, apoptosis, differentiation, and migration, and plays context-dependent roles in human health and disease, particularly cancer and inflammation. Dysregulation of KLF12 expression is implicated in tumor progression, poor prognosis, and susceptibility to inflammatory disorders, making it a potential biomarker and therapeutic target in oncology and immunology. No approved drugs target KLF12 directly, but its pathway involvement suggests future therapeutic interest.
May act via modulation of transcriptional activity, either inhibiting or enhancing key regulatory genes involved in cell cycle and apoptosis (e.g., via the p53/p21 axis). Could affect cancer cell sensitivity to apoptosis-inducing agents.
