Overview

Krueppel-like factor 12 is a zinc finger transcription factor in the Krueppel-like factor family, characterized by three C2H2-type zinc finger motifs at the carboxy-terminal and a variable amino-terminal regulatory domain. It binds GC-rich DNA sequences to regulate gene expression, notably repressing the AP-2 alpha gene by binding to its promoter with the aid of the corepressor CtBP1. KLF12 is involved in cell proliferation, apoptosis, differentiation, and migration, and plays context-dependent roles in human health and disease, particularly cancer and inflammation. Dysregulation of KLF12 expression is implicated in tumor progression, poor prognosis, and susceptibility to inflammatory disorders, making it a potential biomarker and therapeutic target in oncology and immunology. No approved drugs target KLF12 directly, but its pathway involvement suggests future therapeutic interest.
