Overview

Krueppel-like factor 8 (KLF8) is a member of the Sp/KLF family of transcription factors, characterized by a C-terminal DNA-binding domain with three krueppel-like zinc fingers[4][5]. KLF8 functions both as a transcriptional repressor and activator, binding to CACCC-box and GT-box promoter elements. It plays a central role in regulating cell cycle progression at the G1 phase, epithelial-to-mesenchymal transition, and oncogenic transformation[2][5]. KLF8 directly binds androgen receptor (AR) and acts as a novel co-activator, promoting AR target gene transcription, thereby enhancing prostate cancer cell proliferation and tumor growth[1]. Overexpression of KLF8 is associated with several malignancies and cancer progression, making it a potential biomarker for aggressive tumors and a putative therapeutic target in oncology[1][2][4].
