Overview

Krüppel-like factor 16 (KLF16) is a C2H2-type zinc finger transcription factor with affinity for GC-rich DNA sequences. It regulates gene expression in metabolic, endocrine, reproductive, and oncogenic pathways, operating both as a repressor and activator via recruitment of histone deacetylase (Sin3/HDAC) or acetyltransferase (p300/HAT) complexes. Mechanistically, KLF16’s activity and nuclear localization are influenced by phosphorylation, particularly by Src-family kinases. Biologically, KLF16 modulates osteogenic differentiation, is involved in tumorigenesis across several cancers, and serves as a potential prognostic biomarker. It does not have known interacting drugs, but its role as a gene regulator makes it a promising candidate for future therapeutic intervention
