Overview

KxDL motif containing 1 (KXD1) is a small, 20-kDa coiled-coil protein encoded by the KXD1 gene and is a component of the BORC (BLOC-1-related complex) and BLOC-1, essential for lysosomal positioning and biogenesis of lysosome-related organelles such as melanosomes and platelet dense granules. It physically interacts with BLOS1 and other BLOC-1 subunits, and its loss causes mild defects resembling Hermansky–Pudlak syndrome in model organisms. KXD1 is involved in vesicular trafficking, autophagy regulation, and may have disease associations in cancer and psychiatric disorders; however, it is not a known direct therapeutic target.

Mechanism of action

Not a direct therapeutic target; mechanism is via modulation of lysosomal function and trafficking. Altered KXD1 expression may impact responsiveness to nortriptyline, but there is no evidence of direct molecular targeting
