The L-antigen family member 1 isoform A (LAGE-1A) peptide presented by histocompatibility leukocyte antigen A*02:01 is a tumor-associated antigen complex. LAGE-1A is a cancer-testis antigen overexpressed in various cancers with limited expression in normal tissues, primarily the testis. When presented by HLA-A*02:01, a common MHC class I molecule, it forms an epitope recognized by specific T cell receptors. This complex is a target for immunotherapy approaches, including engineered T cell therapies and bispecific antibodies, to redirect T cells to kill tumor cells expressing this antigen. IMCnyeso is an example of such a therapeutic agent. A potential safety concern is off-tumor toxicity due to LAGE-1A expression in the testis.
