Overview

L-Canavanine is a non-proteinogenic amino acid primarily found in leguminous plants such as alfalfa (Medicago sativa), where it serves as a defensive allelochemical against herbivores. [1] It is a structural analog of the essential amino acid L-arginine, distinguished by the replacement of the terminal methylene group with an oxygen atom. [2] This structural similarity allows L-canavanine to act as a potent antimetabolite by competing with L-arginine for transport and enzymatic utilization. [3] A significant biological consequence of this competition is the mistaken incorporation of L-canavanine into nascent polypeptide chains by arginyl-tRNA synthetase, resulting in the production of 'canavanyl-proteins' that are frequently misfolded and dysfunctional. [4] Research has also identified L-canavanine as an inhibitor of nitric oxide synthase (NOS), particularly the inducible isoform (iNOS), suggesting potential therapeutic applications in inflammatory diseases and oncology. [5] However, its clinical development is strictly limited by its well-documented association with the induction or exacerbation of systemic lupus erythematosus (SLE) in humans and primates following the consumption of alfalfa seeds or sprouts. [6]
