Overview

Tyrosinase-related protein 2 peptide antigen (TRP-2) is a peptide epitope derived from the melanocytic enzyme DOPAchrome tautomerase (DCT). TRP-2 is a melanocyte differentiation antigen recognized by cytotoxic T lymphocytes, making it a key target for immunotherapy in melanoma. The peptide antigen is commonly used in peptide-based cancer vaccines to stimulate an anti-tumor immune response. TRP-2 functions biologically as an enzyme in melanin synthesis and is expressed in melanocytes, melanoma, and retinal pigment epithelium, but not in other normal tissues. Immunotherapy approaches leveraging TRP-2 peptide antigens exploit their restricted expression to melanocytic tissues, aiming for tumor selectivity, although there is a risk of autoimmune targeting of normal pigment cells. The molecular target lends itself to investigation as a biomarker for melanoma and as a means of monitoring immune responses in experimental and clinical cancer immunotherapy
