Overview

L-rhamnose isomerase (EC 5.3.1.14) is a bacterial enzyme that catalyzes the reversible conversion of L-rhamnose to L-rhamnulose (UniProt: P0AEE3). It belongs to the isomerase family and typically requires divalent metal ions such as manganese or cobalt for its catalytic activity (PubMed: 11554771). In nature, the enzyme is part of the rhamnose operon, allowing microorganisms to utilize L-rhamnose as a carbon and energy source (PubMed: 25636005). Although it is not a target for current human therapeutic drugs, it is of significant interest in the biotechnology industry for the production of rare sugars like D-allose and L-lyxose (PubMed: 15186101). These rare sugars produced by the enzyme have demonstrated potential medicinal properties, including anti-tumor and anti-inflammatory effects in research settings (PubMed: 18406545). The enzyme's broad substrate specificity makes it a versatile tool for biocatalysis, though it is not directly involved in human disease pathways.
